RT-PCR validation of microarray analysis. Effects of distinct fatty acid (FA) species on ARC expression of ucp3 (A), mcd (B), pdk4 (C), cte1 (D), mte1 (E), adrp (F), and phlda1 (G), as assessed by microarray analysis (i) and real-time RT-PCR (ii). All FA species were conjugated to 1% BSA, and were present at a concentration of 0.4 mM. Control cells were cultured in the absence of FA, but in the presence of 1% BSA. ARCs were challenged for 24 h. Individual gene expression data were initially normalized to the housekeeping gene cyclophilin and are presented as values relative to the control group.
